Mutation detailsRecombineering methods generated a construct in which a cre recombinase cassette was inserted at the transcriptional start site of Tshb in a BAC. Microinjection of the construct generated 4 cre-expressing founders which were crossed to reporter mice for activity analyses. Three lines demonstrated moderate levels of activity in the pituitary gland, but the activity was not specific to thyrotrophs. The other founder line 4 showed higher activity with greater thyrotroph specificity and was selected for further analysis. (J:197591)
