Mutation detailsA vector for recombinase-mediated cassette exchange contained the with the sequence encoding a cre/EGFP fusion protein was inserted into a vector containing the region of Ghrl (from upstream of exon 1 to downstream of exon 5) that had been replaced to make the Ghrltm1Suss LCA allele. The cre/EGFP open reading frame was inserted at the endogenous start codon. The vector was coinjected with a cre expressing plasmid into ES cells carrying the Ghrltm1Suss allele, and the selection cassettes in the LCA allele were replaced with the modified Ghrl sequence. ES cells were injected into blastocysts to generate chimeras that were bred to wild-type Black Swiss mice to obtain germline transmission. These mice were crossed to Tg(ACTBFLPe)9205Dym mice to remove the hygro marker included with the cre/EGFP sequence. (J:195663)
