Mutation detailsThe loxP flanked segment of Mafatm1Eyc, extending from 46 bp upstream of the ATG translation initiation site of the endogenous gene to 23 bp downstream of its polyadenylation signal (and encompassing a FRT-flanked PGK-neo cassette), has been deleted by Cre recombinase. This places the coding sequence for nuclear localized E. coli beta-galactosidase (lacZ), which had followed the 3' loxP site, under transcriptional control of the Mafa regulatory elements. In the developing embryo, cells expressing lacZ were first observed at embryonic day (E)10.5, in the already-aggregated dorsal root ganglia (DRG), where the number of stained cells and intensity of staining increases through E12. Expression is also observed in the trigeminal ganglia from E12.5 and is seen in other parts of the central nervous system, including spinal cord, olfactory bulb, dorsal diencephalon and brain stem.
