Mutation
description
Allele Type:    Targeted (Inserted expressed sequence)
Mutation:    Insertion

Mutation detailsA floxed neo cassette was inserted in intron 13 and a D486A mutation was inserted in exon 14 via homologous recombination. The neo cassette was subsequently removed via in vivo Cre mediated recombination. The D486A mutation creates a kinase inactive protein. Western blot analysis detected a greater than 50 fold reduction in protein expression in homozygous embryos and MEFs. (J:141095)
