• Mutation details: A targeting construct was created that includes exons 1-3 that encode the extracellular domain, a floxed neomycin cassette, a chimeric exon 4 encoding the endogenous transmembrane domain ligated to the CD4 cytoplasmic tail, and a truncated version of the noncoding exon 5. The amino acid sequence at the site of gene fusion is IITLICYHR-CRHQQ with the hyphen representing site of gene fusion. Chimeric mice were mated to Cre expressing transgenic mice to induce germline excision of the neomycin-resistance cassette. Expression of the fusion protein was confirmed by immunoprecipitation with anti-CD8 antibodies. (J:126026)
