Mutation detailsNucleotides -1404 through +61 of the spermatocyte-specific Pgk2 promoter region were joined to the cDNA encoding Cre recombinase followed by a rabbit globin polyadenylation signal. Expression of cre mRNA in testes was demonstrated by RT-PCR analysis. Double transgenic progeny of a cross of mice with this transgene to animals carrying a loxP-flanked EGFP reporter gene exhibit EGFP fluorescence in spermatocytes and spermatids, but not in spermatogonia or somatic cells of the testis. Notes
Two transgenic mice derived from injected oocytes transmitted the transgene to their progeny. The resultant transgenic mouse lines were designated 3 and 24. Because Cre-mediated recombination sometimes failed to occur in double transgenic progeny of line 3, only line 24 was used.
