• Mutation details: The transgene is composed of a cDNA encoding the long form of murine B4galt1, the mouse Mt1 promoter, and human growth hormone 3' polyadenylation sequences. Intron 4 was included in the B4galt1 sequence to increase transgene expression. Expression of long form B4galt1 was verified by RNase protection assay of testes RNA. Transgene expression was also confirmed by enzyme assays, immunoprecipitation enzyme activity, and Western blot analysis. (J:20319)

Notes

Transgenic founders designated 06 and 09 are high-expressing lines and were selected for study. Enzyme activity is present in a variety of tissues examined. and activity is estimated to be 10- to 12-fold higher in testes of transgenic animals compared to wild-type. Induction with zinc increased expression of the transgene only slightly.
