    Locus refinement and candidate gene screening was performed for bone mineral density QTL Bmd1b on mouse Chromosome 1. Subcongenic line analysis was used to localize Bmd1b to a 4 Mb interval between 172 Mb - 176 Mb. Total femur volumetric bone mineral density (vBMD) was measured in 16-week old subcongenic animals. Subcongenic animals carrying CAST/EiJ-derived donor DNA encompassing the Bmd1b critical region on a C57BL/6J genetic background display significantly increased vBMD compared to C57BL/6J controls.

    Expression analysis identified Darc at 173.6 Mb (94 cM) as a potential candidate gene for Bmd1b. Darc expression is 6-fold higher in B6.CAST-Bmd1b subcongenic animals compared to C57BL/6J. Darc<tm1Pogo) homozygous knockout animals exhibit decreased femur vBMD, 13% decreased trabecular bone volume, and 3.8% reduced endosteal circumference. In addition, 26 single nucleotide polymorphisms were identified between CAST/EiJ and C57BL/6J Darc sequences, with 6 SNPs resulting in amino acid substitutions. Thesix non-conservative SNPs were conserved between high BMD phenotype strains CAST/EiJ and C3H/HeJ.
