| Caption | (A) Schematic representation of the wild-type Scrib allele, targeting construct, and mutated Scrib alleles. Excision of the Frt-flanked PGK-Neo cassette in the LoxP-flanked targeting vector was accomplished by crossing Scribfl-Neo mice to Tg(ACTFLPe)9205Dym/0 (Actin-FLPe) mice, producing the Scribtm1.1Phum (Scrib floxed allele (deltaNeo); Scribfl) allele. The insertion of EcoRV and KpnI restriction sites permitted screening for recombinant ES cell clones. Scrib knock-out mice were derived by crossing Scribtm1.1Phum/Scrib+ (Scrib+/fl) mice with a germline Cre-deleter mouse to generate the Scribtm1.2Phum allele (Scrib KO allele).
