immune system
• T cells cultured in vitro under Th2-polarizing conditions including IL-4 keep producing IFN-gamma suggesting a lack of suppression of Th1 differentiation
• T cells cultured in vitro under Th2-polarizing conditions fail to differentiate into Th2 effector cells
• when stimulated in the presence of IL-4, mutant B cells proliferate less and have less surface expression of MHC-II, Thy-1, and CD23 compared to controls
• incubation alone with IL-4 upregulates surface expression of CD23 on wild-type but not mutant B cells
• in response to irradiation despite IL4 treatment
• B cells only have half the rate of proliferation as controls when stimulated in the presence of IL-4
• mice injected with anti-IgD immunoglobulins fail to produce IgE while the same treatment in wild-type mice leads to dramatic increases in IgE
• anti-IgD treated mutant mice have higher levels of IgG2a, IgG2b, and IgG3 than controls
• increases in IgG1 after anti-IgD treatment are similar between mutant and wild-type mice
• B cells express much less MHC-II on their surface when activated in the presence of IL-4
