Limb deformities in Vcantm1.1Hwat/Vcantm1.1Hwat Tg(Prrx1-cre)1Cjt/0 mice

limbs/digits/tail
• at E18.5, metatarsus exhibit an aberrant nodule of hypertrophic chondrocytes surrounded by prehypertrophic and proliferative chondrocytes in a concentric pattern without vascular invasion unlike in wild-type mice
• at E18.5, mice exhibit clefting of the metatarsophalangeal joints compared with wild-type mice
• in the hind limb, mice exhibit distortion of digit morphology compared with wild-type mice
• digits contain a nodule of hypertrophic chondrocytes surrounded by proliferative and prehypertrophic chondrocytes unlike in wild-type mice
• the proximal region of proximal phalanges exhibits a cleft unlike in wild-type mice
• at E14.5, no clear joint interzone stripes are observed unlike in wild-type mice
• at E15.5, digits exhibit a broader interzone of small mesenchymal cells compared to in wild-type mice that gives rise to a wedge-shaped or vertical/longitudinal cavity and lacks hypertrophic chondrocytes
• however, digits in the fore limb are normal
• as early as 1 week of age, especially the proximal phalanges, in the hind limb but not the fore limb
• the joint surface between phalanges is tilted unlike in wild-type mice

skeleton
• as early as 1 week of age, especially the proximal phalanges, in the hind limb but not the fore limb
• the joint surface between phalanges is tilted unlike in wild-type mice
• in micromass culture, chondrocyte differentiation within hind limb digits is delayed compared to in wild-type mice
