growth/size/body
• mice exhibit significantly greater growth retardation than Igfbp4-null mice becoming significant at P20 (78% of normal weight) and continues through week 14

homeostasis/metabolism
• mutants have a 50% reduction in total serum Igf-1 compared to wild-type; Igf-1 bioactivity is significantly suppressed in mutants
• mutants show lower circulating Igf-1 levels than wild-type controls at all time points examined after glucose challenge; percentage increase in Igf-1 levels after challenge is significantly higher in mutants compared to wild-type at 30 minutes (43%) while wild-type only show a 6% increase at 30 minutes and 8% at 60 minutes
• after an overnight fast, mutants have significantly reduced glucose levels vs wild-type mice
• mice show enhanced insulin secretion after glucose challenge
• mutants show significantly decreased glucose levels after glucose challenge (from 30-120 minutes) compared to wild-type or Igfbp3 or Igfbp5 mutants; decrease is significantly enhanced compared to Igfbp4 mutants
• after an overnight fast, mutants have significantly reduced glucose levels vs wild-type mice
• mutants show increased circulating insulin levels after glucose challenge that are significantly higher than the increase seen in wild-type after glucose challenge
• mutants show slightly increased AKT activation levels and significantly decreased Erk/MAPK (72% decreased) activation levels vs wild-type

endocrine/exocrine glands
• islets in mutants have increased area; beta-cell area is increased ~37% compared to wild-type
• mice show enhanced insulin secretion after glucose challenge

adipose tissue
• pads of mutants contain much smaller adipocytes than in wild-type fat pads
• weights of all fat pads are significantly decreased in mutants

muscle
• quadriceps muscle is decreased in weight compared to controls

limbs/digits/tail
• quadriceps muscle is decreased in weight compared to controls
