Mutation details: The transgene was designed by fusing two bacterial artificial chromosomes (BACs) to create an AR BAC construct that includes all eight exons (~180 kbp) of the human androgen receptor gene (AR), ~50 kbp of flanking sequence transcriptionally-upstream of the first AR exon and ~23 kbp of flanking sequence transcriptionally-downstream of the last AR exon. Next, AR exon 1 (which includes the translational start site) was flanked with loxP sites, and a 121 glutamine tract (stabilized with CAA interruptions) was introduced into AR exon 1. No other complete loci exist in the 5' or 3' flanking BAC regions. The founder line 3 identified with 121 polyQ repeats was established.
