Mutation details: A vector containing the following elements was inserted into the three tandem attP sequences the locus previously had been modified with, using phage phiC31: attB site, FRT site flanked STOP cassette, loxP site flanked STOP cassette, CAG promoter, hM3Dq cDNA, sequence coding for T2A self-cleaving peptide, mCherry reporter gene, SV40 poly(A) signal sequences, and second attB site. The hM3Dq sequence is a Gq-coupled human M3 muscarinic DREADD (designer receptor exclusively activated by designer drug). To create the hM3Dq sequence, the wildtype human muscarinic 3 receptor (CHRM3) sequence was modified with two amino acid substitutions (p.Y149C and p.A239G) that abolish receptor affinity for the native ligand acetylcholine (ACh) but allow receptor binding and subsequent activation by the small drug-like molecule clozapine-N-oxide (CNO). The FRT site flanked STOP cassette was removed through subsequent Flp-mediated recombination.
