Mutation detailsSerine codons 316 (TCG), 322 (AGC) and 333 (TCA) in exon 22 (in reference to ENSMUST00000160844) were changed to alanine (GCG, GCC, GCA) (p.S316A, p.S322A, p.S333A) and a loxP and FRT site flanked puromycin resistance gene cassette was inserted into intron 21. The puro cassette was removed through subsequent Cre-mediated recombination. The mutations, in the region of the encoded peptide where heparan sulfate (HS) is added, prevent HS-glycosylation of the affected residues. (J:277096)
