Mutation details: TALEN technology was used to replace the key residues Tyr41, Leu44, and Phe46 of the self-assembly motif with glycine residues: p.Y41G (TAT to GGA), p.L44G (CTT to GGT), and p.F46G (TTT to GGT). These residues correspond to p.Y67G, p.L70G, and p.F72G on build GRCm39. The ssODN was designed with silent mutations within the recognition sequence of Ambn-TALEN to avoid repetitive digestions.
