Mutation details: A construct comprising the tetO tetracycline response element, rabbit beta-globin intron, Yellow Cameleon Nano50, SV40 poly(A) signal sequence and an FRT site flanked neomycin resistance gene cassette, was inserted 799 bp downstream of the Actb poly(A) signal. The neo cassette was removed through subsequent flp-mediated recombination. Yellow Cameleon Nano50 is a Forster resonance energy transfer (FRET)-based genetically encoded Ca2+ indicator (GECI) with a +3 linker (Gly-Gly-Ser) between the calmodulin motif and the M13 peptide. The inclusion of the longer linker segment increases Ca2+ affinity (Kd = 50 nM).
