Mutation detailsMutations (V745L, M761Aand S828A) were introduced into the ATP-binding pocket of the kinase to create an analog sensitive kinase allele and a floxed neo cassette was inserted in intron 17 via homologous recombination. Cre mediated recombination removed the neo cassette. Kinase activity of this allele can be controlled by the administration of 1Na-PP1, a derivative of the general kinase inhibitor PP1. (J:210627)
