Mutation detailsTALEN-targeted homologous recombination corrected the T to G nucleotide substitution in the chocolate mutation (Rab38cht) by reconstituting the wild-type sequence encoding glycine at position 19 and thereby eliminating the exogenous SexAI restriction site (cht-GTT to wt-GGT). An identifying neutral nucleotide replacement in codon 18 (G to A) was used as a unique sequence identifier. Silent exchanges within the TALEN binding region remove an endogenous ApaLI site. This allele was identified in a line established from founder Rw2. (J:205086)
