Mutation details: The CAG promoter drives ubiquitous expression of reverse tetracycline transactivator 3 with two mutations: 1) modified to bind to the tetracycline responsive element in the presence of doxycycline and 2) three mutations within the transactivator (S12G, F86Y, and A209T) that increase its sensitivity to doxycycline by 25-fold over the original rtTA without increasing background activity.
