Mutation detailsOne transgene construct consists of human HLA-DRA*0101 and HLA- DRB1*0401 (Gly-86) cDNAs, each placed downstream of the histocompatibility 2, class II antigen E alpha promoter and the rabbit beta-globin intron and upstream of the beta-globin polyadenylylation site. The other transgene construct is composed of the mouse CD3 antigen delta polypeptide promoter followed by a human CD4 cDNA fragment encoding exons 2-4 and part of 5, a genomic fragment encoding the remainder of CD4 exon 5, exons 6-9, and part of exon 10, and SV40 polyadenylation signal sequences. The two constructs were co-microinjected into single-cell embryos. Five triple-transgenic lines were established, of which one was chosen for further study. The chosen line has approximately five copies of the HLA construct and two or three copies of the CD4 construct, with both constructs in strong linkage disequilibrium. Flow cytometry experiments showed expression of the human transgenes in peripheral blood mononuclear cells. Mice with this allele express the HLA-DR4 human serotype. (J:265804)
