Mutation detailsA vector containing the following elements was injected into pronuclei of the host embryos: mouse promoter, Kozak sequence, the mouse cDNA, 29 bp Myc tag, two stop codons, Kozak sequence and the human growth hormone 1 minigene. Zygotes were isolated from BTBR (Black and Tan Brachyury) females. BTBR mice are models for the study of type 2 diabetes (T2DM) and autism spectrum disorder (ASD) and carry the Disc1del, Itpr3tf and T+ alleles. The human gene confers high levels of expression of this allele. Expression of the mouse gene was confirmed by RT-PCR, but unintended expression of the human gene was also shown. Likewise, both the mouse and human proteins were detected. The pound symbol (#) is used when no line is specified. (J:227290)
