Mutation detailsFertilized mouse eggs were coinjected with mouse immunoglobulin heavy and light chain transgene constructs whose rearranged heavy chain VDJ segments and kappa light chain VJ segments were derived from the HyHEL10 plasmacytoma cell line, which produces a high affinity antibody to chicken lysozyme (commonly called hen egg lysozyme (HEL)). The heavy chain variable segments are joined to a genomic DNA segment from a BALB/c-derived plasmacytoma (Igha allotype) containing the Ighm switch region, the first four exons exons encoding the IGH-mu constant region, a chimeric Ighm/Ighd M1 exon, and the Ighm M2 exon and 3' flanking DNA. In the M1 exon, which encodes the extracellular spacer, the transmembrane domain and the short cytoplasmic tail, the sequence encoding the IGH-Cmu extracellular spacer has been replaced by that from Ighd. The light chain variable segments are joined to a genomic DNA segment containing the Igk constant region and 3' flanking DNA of the Igka allotype. Each construct is expressed from the promoter upstream of the leader sequence of the V-region segment. (J:78308, J:104190, J:211176)
