Mutation detailsA fragment encoding the cre recombinase fused to a mutated ligand-binding domain of the estrogen receptor (ERT2) was cloned downstream of the Cd4 gene promoter/enhancer/silencer to generate the transgene construct. The construct also contained the human growth hormone (hGH) polyadenylation sequence. Pronuclei of fertilized eggs were microinjected with the purified construct. Founder lines 11 and 19 were established. These were crossed to R26R-EYFP reporter mice. CD4 T cells were isolated from tamoxifen-treated double mutant animals and 50 and 30% of cells analyzed by FACS from lines 11 and 19 respectively showed EYFP expression. Line 11 had greater recombinase activity and was used for further analyses. (J:191897)
