Mutation detailsA targeting vector was designed to replace the entire coding region of the achaete-scute complex homolog 1 (Ascl1 or Mash1) with a CreERT2 fusion protein and a frt-flanked neomycin (neo) selection cassette. The construct was electroporated into 129S/SvEv-derived embryonic stem (ES) cells. Correctly targeted ES cells were injected into C57BL/6 blastocysts and the resulting chimeric mice were bred to C57BL/6 mice. These mice were then bred to mice containing Flp-recombinase to remove the Frt-flanked sequence. These mice were then backcrossed at least 5 generations to and outbred ICR strain generate a colony of Ascl1-CreERT2 mice. (J:171439)
