Molecular description:

The targeting vector was designed to insert Cre/ERT2 cassette, polyA tail, and loxP-flanked PGK-Neo cassette in the opposite transcriptional orientation to the first exon of the Ndrg1 gene at the position of the initiation codon. The knock-in allele disrupted the first exon, thereby deleting the expression of Ndrg1.
