Mutation details: Human TCR and TCR variable Ag recognition sequences isolated from a T1D donor (TCR 6H9, recognizing insulin B9-23 in a DQ8 (HLA-DQA*0301, DQB*0302) or DQ8 trans-restricted) fashion were fused to murine constant regions to create chimeric TCR genes. TCRalpha sequences were inserted into vector pCD2, which drives expression from a human CD2 promoter. TCRbeta sequences were inserted into p428, which drives expression from a murine CD4 promoter. Transgenic fragments were isolated and injected into NOD-cMHCI/II-/- (NOD/ShiLtDvs-H2-K1d-em1Dvs H2-Ab1g7-em1Dvs H2-D1b-em5Dvs/DvsJ) zygotes. The integration site and the copy number of the HLA and TCR transgenes are unknown. This is founder line 2.
