Mutation details: This transgene comprises a 1,385-bp mouse genomic DNA fragment containing an enhancer sequence residing ~3 kb upstream of the coding sequence for atonal bHLH transcription factor 1 (Atoh1, also called Math1; GenBank: AF218258, nt 1-1385) followed by the human beta-globin basal promoter (nt -31 through +12) driving expression of the full-length, 3.1-kb mouse Bmi1 polycomb ring finger oncogene (Bmi1) cDNA. Two transgenic lines (of four generated), designated T25 and T26, overexpressed Bmi1 mRNA, as determined by quantitative reverse transcription-polymerase chain reaction (qRT-PCR) analysis, and BMI1 protein, as quantified by immunoblot analysis, in cerebellum; immunohistochemical labeling of cerebella of 6-day-old mice localized expression of BMI1 to the outer layer of the external granule cell layer, where proliferation of granule cell precursors occurs.
