Mutation detailsThis transgene comprises a single, stably integrated copy of a pTRIP deltaU3-derived lentiviral vector containing the coding sequences for green flourescent protein (GFP) and mouse FK506 binding protein 11 (FKBP11, also called FKBP19) under transcriptional control of the human ubiquitin C (UBC) gene promoter. A DNA segment between the GFP and Fbkb11 sequences that encodes a "self-cleaving" picornavirus T2A peptide results in stoichiometric coexpression of the two proteins. Quantitative real-time RT-PCR demonstrated overexpression of Fkbp11 mRNA in unsorted bone marrow cells, splenocytes, isolated splenic B cells and unsorted lymph node cells from Fkbp11high transgenic mice. Immmunoblot analysis verified overexpression of the FKBP11 protein in splenocytes, splenic sorted B cells and lymph node cells, but the protein was barely detectable in bone marrow cells. (J:227176)
