Mutation detailsThe RP23-92L23 bacterial artificial chromosome (BAC) containing mouse Ins2 was modified by first replacing the Ins2 coding region by galK. Then the galK insert was replaced by the human diphtheria toxin receptor (HBEGF) cDNA. This results in the human diphtheria toxin receptor under the control of the mouse Ins2 promoter. Lines 5, 14, 17, and 33 were generated. Transgene expression is seen in pancreatic beta cells and administration of diphtheria toxin results in HBEGF expressing cells. (J:204420)
