Mutation details: This transgene contains a tetracycline response element, comprising seven copies of the 42-bp tet operator sequence, between two minimal cytomegalovirus promoters, one driving expression of the E. coli lacZ (beta-galactosidase) gene and the other of a cDNA encoding mouse secreted phosphoprotein 1 (also called osteopontin) (nt 9-955, GenBank accession BC057858). Both genes are inactive except in the presence of a second transgene expressing the tetracycline transactivator (tTA) or reverse transactivator (rtTA) protein, which permits regulation of their expression by administration/discontinuation of doxycycline.
