Mutation detailsThis transgene contains a cDNA encoding the short form of human CASP8 and FADD-like apoptosis regulator (CFLAR; also called FLIP) with an N-terminal FLAG tag inserted into the p1017 expression vector, which contains a 3.2 kb fragment of the mouse lymphocyte protein tyrosine kinase (Lck) gene that includes the thymocyte-specific proximal promoter through nucleotide +37 (relative to the transcription initiation site) upstream of the nearly full-length, 5' truncated human growth hormone (GH1) gene; the insertion site is in the truncated first exon of GH1, which encodes the 5' UTR. Western blot analysis demonstrates high levels of expression in thymocytes and splenic T cells of transgenic mice. (J:97825)
