Mutation detailsThis transgene contains the bovine keratin 5 promoter driving expression of a cDNA encoding the long isoform of mouse transformation related protein 63, which includes the amino-terminal transactivation domain ("TAp63"), with a hemagglutinin (HA) tag at its amino terminus. Immunoblot analysis using antibodies vs. either TRP63 or HA demonstrates expression specifically in tissues where keratin 5 normally is expressed, including eye, lung, thymus and the basal layer of the epidermis. Immunostaining of skin biopsies and of cultured primary keratinocytes from transgenic mice showed that the transgene is overexpressed as a nuclear protein in cells of the epidermal basal layer.
