Mutation detailsThe transgene contains the cDNA encoding human myeloid cell leukemia sequence 1 (BCL2-related) joined to the promoter and 5' regulatory sequences of the human eukaryotic translation elongation factor 1 gene, including 1.5 kb of 5' flanking DNA, the first (noncoding) exon and the first intron. RT-PCR analysis detected expression in all organs/tissues examined; levels of expression differed among organs, being greatest in the islets of Langerhans, but was much lower everywhere than that of the endogenous gene. Levels in all tissues were too low for detection by northern blot analysis. (J:83768)

Notes
Three transgenic lines were created: E2, estimated to have 20 copies of the transgene and an expression level in the islets of Langerhans ~3.5-fold that of beta-actin; E3, with 50 copies and an islet expression level ~0.5-fold that of beta-actin; and E12, with 20 copies and about 0.75-fold the beta-actin expression level. Mice of lines E2 and E12 present a similar histological phenotype which appears to be absent in line E3 mice.
