Notes
  • Experiment
To obtain a double transgenic population, E(mu)Pim1 transgenic mice were crossed with animals from three different E(mu) L-Myc transgenic lines (EL27.3, E35.6 and EL31.14). L-Myc/Pim1 mice infected with Moloney Murine Leukemia virus (MoMuLV) were shown to be twice as susceptible to tumors than non-infected mice. DNA extracted from these animals tumor tissue was then hybridized with several probes for known proviral integration sites. The most proviral integrations (20 of 58 tumors) were found to have occurred in the Pal1 region of the Gfi1 locus. Inverse PCR reactions to clone genomic sequences flanking proviral integration sites revealed mouse sequences bearing homology to rat Gfi1 cDNA. A full-length Gfi1 cDNA probe confirmed several more cases of integration at this site in other tumors originating in the transgenic strain.
