  • Experiment
The authors created two subcongenic mouse strains, D2.B6-Mop2-P1 and D2.B6-Mop2-D1, that
break the previously Mop2 QTL interval (28.8 Mbp in
D2.B6-Mop2) into two segments (9.5 Mbp in D2.B6-Mop2-P1 and 17.2 Mbp in
D2.B6-Mop2-D1). D2.B6-Mop2-P1 congenic mice exhibit a strong preference for
morphine over quinine, whereas D2.B6-Mop2-D1 congenic mice avoid morphine (similar to DBA/2J mice).

The subcongenic mice, D2.B6-Mop2-P1 and D2.B6-Mop2-D1 can be can be divided into two segments, with the one breakpoint between markers D10Mit28 and D10Mit211 Mapping of chromosome 10 genes within the region of the breakpoint indicate that both D2.B6-Mop2-P1 and D2.B6-Mop2-D1 mice were homozygous for the D2 allele at the Grm1 and Stxbp5 loci.

Bascially the D2.B6-Mop2-P1 subcongenic strain contains C57BL/6J sequences from the centromere to D10Mit28 , whereas D2.B6-Mop2-D1 congenic mice contain C57BL/6J sequence between marker D10Mit211 and marker D10Mit51. These QTL segments have been labeled Mop2a and Mop2b respectively.

The characterization of these congenics reaffirms the mapping of the Mop2 QTL to mouse Chromosome 10.
