An X-Chromosome enriched library was contructed by flow sorting mitotic chromosomes from a SV40 cell line derived from a 22CD feral mouse. In 22CD mice all chromosomes except 19 and X are fused in a metacentric arrangement. Total mouse DNA was used as a probe to identify recombinant phages from this library devoid of repetitive sequences. A selection process was employed using plaque lift hybridization followed by Southern blotting. Repetitive negative inserts identified by the process were used to probegenomic blots of EcoRI digested DNA from Chinese hamster x mouse somatic cell hybrids. Hybridization was detected in three hybrid lines containing a complete X Chromosome, and was not detected in hybrids lacking an X. Five inserts were X Chromosome specific. These were 45, 52, 66, 87 and 100 representative of loci DXPas1, DXPas2, DXPas3, DXPas4 and DXPas5 respectively.
