Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1Jand (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fatdiet compared to DBA/1J males.

Suggestive linkage to plasma triglyceride levels on a high fat diet for 8 weeks mapped to 31 cM on mouse Chromosome 15 with LOD=3.8. This locus has been detected in other mouse crosses and was designated Tgq27 (triglyceride QTL 27).
