Linkage analysis was performed using a backcross derived from closely related inbred strains DBA/1J and DBA/2J. The genetic composition of these 2 inbred strains differs by just 5.6% at the SNP level. A total of 294 mice from (DBA/1J x DBA/2J)F1 x DBA/1Jand (DBA/1J x DBA/2J)F1 x DBA/2J populations were screened for 6,949 polymorphic SNP markers. Backcross mice were placed on a high-fat diet at 8 weeks of age and phenotyped for plasma triglyceride and HDL concentrations at 8, 11 and 16 weeks of age. Parental strain DBA/2J displays significantly increased plasma triglyceride concentrations on a CHOW diet compared to parental strain DBA/1J. Male DBA/2J animals display significantly increased plasma triglyceride and HDL concentrations after an 8 week high fat diet compared to DBA/1J males.

Linkage to triglyceride concentrations on a CHOW diet mapped to 18 cM on mouse Chromosome 3 at peak marker rs3700620 (Tgq13; LOD=4.7) and 71 cM at rs3668158 (Tgq15; LOD=5.2). DBA/2J-derived alleles at Tgq13 and Tgq15 confer increased triglycerides on a CHOW diet. The confidence interval for Tgq13 spans 10 cM - 74 cM and the confidence interval for Tgq15 spans 18 cM - 71 cM.
Linkage to plasma triglycerides in males after a 3 week high fat diet mapped to 41 cM at rs3668158(136.27 Mb) with LOD=4.8. This locus is named Tgq14 (triglyceride QTL 14). DBA/2J-derived alleles at Tgq14 confer increased triglycerides in males. The Tgq14 confidence interval spans 14 cM - 68 cM. [Table 3].

Linkage to plasma HDL after 3 week highfat diet mapped to 2cM on Chromosome 3 with LOD=4 and is named Hdlq61 (HDL QTL 61). [Table 2].
