Linkage analysis was performed on 1040 (LG/J x SM/J)F2 animals to identify genetic loci associated with skeletal traits. The F2 population consists of two sets of intercrosses where intercross I (n=533) was phenotyped at 10-weeks of age and intercross II(n=507) was phenotyped between 72- and 237-days of age. Parental strain LG/J displays increased body weight compared to SM/J. Seventy skeletal traits were measured. Intercross I was genotyped for 76 microsatellite markers on 19 autosomes (excluding X chromosome). Intercross II was genotyped at 95 microsatellite markers and 369 SNP markers, including 16 SNPs on chromosome X. One hundred five pleiotropic loci were identified representing 798 single trait QTLs. The average number of traits associated per QTLwas 7.6. In general, LG/J-derived alleles confer increased skeletal trait measurements.

    On mouse Chromosome 19, four skeletal trait QTLs mapped to 0 cM (Plskt89; LPR=3.51), 12.9 cM (Plskt90; LPR=3.83), 33 cM (Plskt91; LPR=5.18) and 56.2 cM (Plskt92; LPR=1.98).

Plskt89 (pleiotropic skeletal traits 89) maps to 0 cM and is associated with hindlimb obturator foramen length and first lumbar length in females. Plskt89 is also associated with pubis length (both sexes). The QTL confidence interval spans 0 cMto7 cM.

Plskt90 (pleiotropic skeletal traits 90) maps to 12.9 cM and is associated with five skeletal measurements. The Plskt90 QTL interval spans 9 cM to 18 cM. This locus is linked to femur length, sacral length, ulna length and third caudal length in females. Plskt90 also displays linkage to mandible square root of masseteric area in both sexes.

Plskt91 (pleiotropic skeletal traits 91) is located at 33 cM. This locus is linked to skull frontal bone length in males, forelimb superior spinous length andcranial vault length. The QTL confidence interval spans 26 cM to 43 cM.

Plskt92 (pleiotropic skeletal traits 92) is located at 56.2 cM. This locus is associated with eighth caudal length in females and forelimb infraspinous length. The Plskt92 confidence interval spans 38 cM to 71 cM.
