A QTL for total food volume intake mapped to 14 cM on mouse Chromosome 17 near D17Mit198 (LOD=7.6). This locus was mapped in an existing cross of 502 (C57BL/6J x CAST/EiJ)F2 animals and is designated Tfv1 (total food volume 1). The Tfv1 interval spans 10cM - 26 cM. F2 animals were assessed at 7-10 weeks of age for food intake. Genome scan was performed using 98 polymorphic markers at a 20 cM resolution. Parental strain CAST/EiJ consumes more food compared to parental strain C57BL/6J over the same period of time.

Tfv1 was confirmed using a congenic line carrying CAST/EiJ-derived DNA on chromosome 17 from 3 cM (D17Mit19) to 37.8 cM (D17Mit91) on a C57BL/6J genetic background. Congenic animals displayed higher consumed food volume over a 10-day period compared to C57BL/6J control animals. Congenic animals also displayed a higher rate of gastric emptying compared to C57BL/6J controls.

The Tfv1 interval coincides with previously identified QTLs Mnic1 (20 cM) and Kcal2 (20 cM). Potential candidate gene Glp1r (18 cM) maps to this interval and displays decreased gene expression in congenic animals compared to C57BL/6J controls.
