  • Experiment
Linkage analysis was performed on 90 animals from a C57BL/6J x (C57BL/6J x LP/J)F1 backcross to identify QTLs associated with isoflurane minimum alveolar concentration (MAC). A panel of 65 polymorphic markers was used for genome scan. Parental strain LP/J displays higher isoflurane MAC compared to parental strain C57BL/6J. The highest linkage was detected at 15 cM on mouse Chromosome 7 near D7Mi246 (LOD=1.7), corresponding to 30 Mb. C57BL/6J-derived alleles confer increased isoflurane MAC at this locus. SNP analysis of 19 inbred strains confirmed association of isoflurane MAC with the proximal chromosome 7 QTL. This locus is named Imacq (isoflurane minimum alveolar concentration QTL). Previously identified QTL mapping to chromosome 7 include Lorp1 (40 cM) and Etan1 (29 cM).
