    Previously identified QTL Sicd1 (seizure-induced cell death 1) on distal mouse Chromosome 18 was confirmed and refined using congenic line analysis. C57BL/6J-derived donor DNA between D18Mit141 (45 cM) and D18Mit25 (57 cM) was introgressed onto a FVB/NJ genetic background for six generations to create FVB.B6-Sicd1 mice. Parental strain FVB/NJ is susceptible to kainic acid seizure induced neuronal cell death whereas parental strain C57BL/6J is resistant. As expected, FVB.B6-Sicd1 animals were resistant toseizure-induced neuronal cell death, especially in area CA3, thus confirming the presence of Sicd1 on distal chromosome 18. Interestingly, FVB.B6-Sicd1 congenics display decreased latency to kainic acid-induced seizures, which suggests that the Sicd1 locus protects against neuronal death, but not seizure susceptibility.

    A series of four overlapping sub-congenic lines were derived from FVB.B6-Sicd1. Significant resistance to seizure-induced neuronal death in one sub-congenic line narrowed the Sicd1 interval to a 2 cM region between D18Mit129 (55 cM) and D18Mit25 (57 cM). A potential candidate gene in this interval is Galr1 (55 cM).
