Experiment
Wg2 (weight gain in high growth mice 2) and Wg4 (weight gain in high growth mice 4) were previously mapped to mouse Chromosomes 2 and 11, respectively. These loci were investigated further by construction of speed congenic lines carrying C57BL/6J-Socs2hg-derived DNA at Wg2 or Wg4 on a CAST/EiJ genetic background. These lines are designated HG.CAST-(D2Mit329-D2Mit457) for the Wg2 congenic and HG.CAST-(D11Mit260-D11Mit255) for the Wg4 congenic. Parental strain C57BL/6J-Socs2hg exhibits significantly increased growth trajectory after 3 weeks of age compared to CAST/EiJ. Over 700 F2 animals were produced from [C57BL/6J-Socs2hg x HG.CAST-(D2Mit329-D2Mit457)]F2 and [C57BL/6J-Socs2hg x HG.CAST-(D11Mit260-D11Mit255)]F2 intercrosses. Animals were analyzedat 9 weeks of age and screened for nineteen polymorphic markers on chromosome 2 (average resolution = 2.7 cM) and eleven markers on chromosome 11 (average resolution = 3 cM). Several novel body trait QTLs were identified and are described below.
On mouseChromosome 2, linkage to 6-week and 9-week body weight mapped to 9 cM (112 Mb) near D2Mit207 and is named Wg5 (LOD=9 and 10.8, respectively). The Wg5 confidence interval spans 2 cM - 29.5 cM. In males, Wg6 is associated with 6-week and 9-week body weightat 27.5 cM (150 Mb) near D2Mit260 (LOD=7.5 and 6, respectively). CAST/EiJ-derived alleles at Wg5 and Wg6 confer decreased body weight.
Linkage to spleen weight mapped to 30 cM near D2Mit262 (Swq6, LOD=11). CAST/EiJ-derived alleles at Swq6 confer increased spleen weight with additive inheritance in males, but with dominant inheritance in females. The Swq6 confidence interval spans 26 cM - 42 cM. Swq6 accounts for 16.7% of the variance.
Linkage to kidney weight mapped to 12.5 cM near D2Mit17 (Kwq13, LOD=9.8 in males) and 41 cM (Kwq14, LOD=7.9). CAST/EiJ-derived alleles at Kwq13 confer increased kidney weight with additive inheritance in females, while heterozygous male mice have increased kidney weight. Kwq13 explains 15% of the variance and the confidence spans 8.5 cM - 13.8 cM. Kwq14 explains 12.3% of the variance.
Linkage to tail length in females, Tailq7, mapped to 12.5 cM (120 Mb) near D2Mit17 (LOD=6.7). This locus explains 10.5% of the variance. CAST/EiJ-derived alleles at Tailq7 confer decreased tail length in females with an additive mode of inheritance. The Tailq7 confidence interval spans 2.5 cM - 22.5cM.
Linkage to body length, Bdlnq7, mapped to 33 cM (159 Mb) near D2Mit196 (LOD=5.3). Homozygosity for CAST/EiJ-derived alleles at Bdlnq7 confersdecreased nasal-to-anal length with dominant inheritance and the effect is stronger in females. This locus explains 8.4% of the variance. The confidence interval spans 0.5 cM - 37.5 cM.
Total fat pad mass, Fatmq1 and Fatmq2, mapped to 20.3 cM (LOD=8.7near D2Mit223) and 37 cM (LOD=5.3), respectively. CAST/EiJ-derived alleles at Fatmq1 confer decreased total fat mass in females with additive inheritance. Fatmq1 explains 13.5% of the variance and the confidence interval spans 9.7 cM - 25 cM. CAST/EiJ-derived alleles at Fatmq2 decrease total fat mass in both males and females. Fatmq2 locus explains 8.5% of the variance.
Linkage to plasma total cholesterol, Tcq1 and Tcq2, mapped to 16.5 cM (LOD=5.8, 9.2% of variance) and 32 cM near D2Mit262 (LOD=6.8, 10.7% of variance), respectively. CAST/EiJ-derived alleles at Tcq1 and Tcq2 confer decreased plasma total cholesterol. The Tcq2 confidence interval spans 12.5 cM - 44.5 cM.
