    The am3 locus was narrowed to a 4.4 cM interval between D3Mit13 and D3Mit125 by the construction and analysis of several sub-congenic lines. Differing lengths of donor DNA from inbred strain MSM/Ms spanning the am3 locus were introgressed onto the genetic background of EL/Seac. Parental strain EL/Seac has a high incidence of absence of M3 molars whereas MSM/Ms does not have absent M3 molars. Congenic animals displayed significantly decreased incidence of absent M3 molars.

    Lef1 and Egf are candidate genes mapping to the am3 interval. Sequence analysis of Lef1 and Egf did not reveal any sequence polymorphisms in the coding region between EL/Seac and MSM/Ms strains. This result suggests that neither candidate gene is likely to be responsible for am3. However, the introns and regulatory sequences of Lef1 and Egf have not yet been analyzed.
