A previously identified QTL named Scpro5 on mouse Chromosome 2 was confirmed and further characterized in this study using a congenic line. Scpro5 maps to approximately 73 cM and is associated with early hematopoietic progenitor cell numbers and hematopoietic stem cell numbers in aged animals. A congenic was constructed which carries DBA/2J-derived donor DNA from 130 Mb to 180 Mb on a C57BL/6J genetic background. This interval contains Scpro5 and its entire 95% confidence interval. Parental strain C57BL/6J does not show decline in hematopoietic stem cell numbers with increasing age whereas hematopoietic stem cells numbers in DBA/2J decreases after 1 year of age. Congenic animals display a phenotype similar to the DBA/2J donor strain. 24 month congenics have 2-fold less hematopoietic stem cells compared to age-matched C57BL/6J controls. This result confirms the existence of the stem cell aging QTL Scpro5 on mouse Chromosome 2.
