    Linkage analysis was performed on a population of (129-Trp53tm1Tyj/J x CE/J)F2 intercross animals to identify modifiers of Trp53. Ratio distortion was observed in F2 animals with a significant decrease in the number of Trp53tm1Tyj heterozygous and homozygous animals. A locus named Motp1 (modifier of Trp53 1) mapped to 42 cM on mouse Chromosome 11 near D11Mit90 with LOD=11.2. This locus is very close to the Trp53 locus (39 cM). Motp1 is a recessive modifier derived from the CE/J genetic background. Homozygosity for CE/J-derived alleles at Motp1 in conjunction with heterozygosity or homozygosity for Trp53tm1Tyj confers increased embryonic lethality. Motp1 was also detected using backcross and congenic analysis, thus verifying its presence.

    Suggestive linkage to Trp53-dependent embryonic lethality was also detected at 35 cM on mouse Chromosome 6 near D6Mit9 (LOD=1.44) and at 38 cM on mouse Chromosome 16 near D16Mit5 (LOD=1.65).
