Animals congenic for the Hyplip2 locus were created by introgressing an MRL/MpJ-derived region between D15Mit176 (10.6 cM) and D15Mit16 (61 cM), which contains the Hyplip2 region, onto a BALB/cJ genetic background. On a CHOW diet, the congenic animals exhibit increased triglycerides and plasma cholesterol levels compared to background strain BALB/cJ. On an 8-week atherogenic diet congenic animals exhibit a 30-fold increase in atherosclerotic lesions and a 2-fold decrease in HDL cholesterol levels compared to background strain BALB/cJ. Heterozygous animals exhibit an intermediate phenotype, suggesting a dominant mode of inheritance for MRL/MpJ allele of Hyplip2.

The Hyplip2 region was narrowed to a 30 cM interval in the distal portion of the QTL by subcongenic strain analysis. Potential candidate genes mapping near the Hyplip2 peak are Ly6 (42.7 cM) and Fbxo32 (atrogin-1). Hyplip2 maps near a previously identified cholesterol absorption QTL named Chab4 (58.1 cM).
