An RFLP was detected in the C1qa upstream region between inbred strains NZB, NZW, and BALB/c. NZB and (NZB x NZW)F1 hybrid animals exhibit decreased serum C1qa activity compared to NZW animals. Authors speculate this polymorphism may have a correlation to systemic lupyus erythematosus (SLE) suscecptibility.

Genome scan was performed on a population of NZB x (NZB x NZW)F1 backcross animals to assess linkage of the RFLP to SLE traits. Linkage to low serum C1q production was detected at D4Mit70 (62 cM) on mouse Chromosome 4 with LOD=3.6. This locus maps very close to C1qa at 66 cM. Linkage to proteinuria was also observed at D4Mit70 with LOD=5.2.

The H2 locus on mouse Chromosome 17 showed significant linkage to proteinuria with LOD=17.5. This locus appears to interact with the D4Mit70 locus to influence susceptibility to SLE. Animals heterozygous for NZB/NZW alleles at D4Mit70 and carring H2d/z alleles exhibit earlier onset and higher incidence of proteinuria compared to all other genotype combinations.
