Linkage analysis was performed on 3 different BSB (B=C57BL/6J; S=SPRET/Ei) backcross populations to identify QTLs associated with obesity traits. The first backcross population consists of 361 (B6.129P2-Lipctm1Unc x SPRET/Ei)F1 x B6.129P2-Lipctm1Unc animals. The second backcross population consists of 199 (B6.129P2-Lipctm1Unc x SPRET/Ei)F1 x C57BL/6J animals. The third backcross population consists of 183 (C57BL/6J x SPRET/Ei)F1 x B6.129P2-Lipctm1Unc animals. At approximately 5 months of age the animals were phenotyped and QTLs were identified using epistatic and non-epsistatic models.

A previously identified QTL, Bsbob on mouse Chromosome 2, was detected in this study. Bsbob maps to 81.7 cM near D2Mit109 and is linked to percent body fat and adiposity index. SPRET/Ei alleles confer decreased obesity (leanness) at Bsbob.

A QTL on central mouse Chromosome 7 mapped between D7Mit8 (60 cM) and D7Mit103 (63.5 cM) and is named Bsbob2. SPRET/Ei alleles confer increased obesity traits (increased body weight, increased fat mass, increase fat percentage) at this locus. Bsbob2 maps near a previously identified obesity QTL named Adip3 (46 cM).

Several interacting QTL pairs were detected on chromosomes 6, 12, and 15. These loci are named Bsbob3, Bsbob4, and Bsbob5, respectively.

Bsbob3 maps to 26.5 cM (D6Mit355) on mouse Chromosome 6 and is observed to interact with Bsbob4 at 52 cM (D12Mit27) on mouse Chromosome 12 to influence total cholesterol levels. SPRET/Ei alleles confer increased total cholesterolfor this locus pair.

Bsbob4 is observed to interact with Bsbob5 at 20.2 cM (D15Mit152) on mouse Chromosome 15 to influence percent body fat. SPRET/Ei alleles confer decreased obesity at Bsbob5.

Bsbob (chromosome 2) is observed to interact with Bsbob4 (chromosome 12) to influence percent body fat, adiposity index, total fat mass, and body weight. SPRET/Ei-derived alleles confer increased obesity traits at Bsbob4.
